Introduction: This study aimed to investigate the effects of 8 weeks of aerobic training and cannabidiol (CBD) supplementation on beta-cell function and pancreatic tissue structure in rats fed a high-fat diet (HFD).
Methods: This experimental study used 32 male Wistar rats (8 weeks old). After 8 weeks of HFD, the rats were randomly divided into 4 groups: HFD, HFD+CBD, HFD+AE, and HFD+CBD+AE. The aerobic exercise program consisted of 5 sessions per week for 8 weeks on a treadmill for rodents. Rats in the supplement group received CBD at a dose of 10 mg/kg, either 2 hours after the training session on training days or at the same time on non-training days. Finally, pancreatic tissue was collected for histological examination.
Results: All therapeutic interventions, including CBD, AE, and CBD+AE, significantly reduced serum glucose, compared to the HFD group. However, insulin levels increased in the HFD+CBD group, compared to the HFD group (P=0.0281). The HOMA-IR index showed a significant decrease only in the HFD+AE group, compared to the HFD group (P=0.0342). Conversely, the HOMA-Beta index showed a significant increase only in the HFD+CBD (P=0.0002) and HFD+CBD+AE (P=0.0017) groups, compared to the HFD group.
Conclusion: These results suggest that although each of these interventions is beneficial on its own, the combination of aerobic exercise and cannabidiol supplementation may represent a more comprehensive and powerful therapeutic strategy to address pancreatic damage and metabolic disorders associated with obesity and diabetes.
